چکیده

In this study, we reported for the first time that a new compound named 3113, which is the dipeptidomimetic of paminobenzoic acid showed multi-drug resistance (MDR) reversal activity. In MDR cancer cells, no matter acquired or inherent, combination with 3113 enhanced doxorubicin (DXR) cytotoxicity by 8 to 10 folds, which was slightly higher than the effect of verapamil (VPL). 3113 was indicated to enhance the cytotoxicity of anticancer drugs through increasing its intracellular accumulation in a time-dependent manner. According to the result of FCM (flow cytometry), the intracellular fluorescence representing the concentration of intracellular DXR was improved by 10 folds as compared with the MDR cells without 3113. Western blotting analysis of P-glycoprotein (P-gp) confirmed that 3113 can alter P-gp expression in a time-dependent way. In conclusion, MDR reversal activity of 3113 depends on, at least in part, the inhibition of P-gp expression.

CRISPR/Cas9, a new approach to successful knockdown of ABCB1/P-glycoprotein and reversal of chemosensitivity in human epithelial ovarian cancer cell line

Objective(s): Multidrug resistance (MDR) is a major obstacle in the successful chemotherapy of ovarian cancer. Inhibition of P-glycoprotein (P-gp), a member of ATP-binding cassette (ABC) transporters, is a well-known strategy to overcome MDR in cancer. The aim of this study was to investigate the efficiency and ability of CRISPR/Cas9 genome editing technology to knockdown ABCB1 gene expression ...

RNAi Induced Inhibition of MRP1 Expression and Reversal of Drug Resistance in Human Promyelocytic HL60 Cell Line

Multidrug resistance (MDR) is a complex phenomenon in which many different genes regulating drug transport, cellular repair, detoxification and drug metabolism are involved. Nevertheless, in most drug resistant cell lines and cancer patients up-regulation of ABC-transporter genes such as MDR associated Protein (MRP1) gene could be at the basis of the drug resistance phenotype. We aimed to decre...

Design real-time reversal of tumor multidrug resistance cleverly with shortened carbon nanotubes

Multidrug resistance (MDR) in tumors renders many currently available chemotherapeutic drugs ineffective. Research in nanobiotechnology-based therapeutic alternatives has provided innovative and promising strategies to overcome MDR. The aim of this study was to investigate whether the new strategy of a co-loaded reversal agent and chemotherapeutic drug with shortened carbon nanotubes (CNTs) wou...

Lamellarin O, a Pyrrole Alkaloid from an Australian Marine Sponge, Ianthella sp., Reverses BCRP Mediated Drug Resistance in Cancer Cells

ATP binding cassette (ABC) transporters, such as P-gp, BCRP and MRP1, can increase efflux of clinical chemotherapeutic agents and lead to multi-drug resistance (MDR) in cancer cells. While the discovery and development of clinically useful inhibitors has proved elusive to date, this molecular target nevertheless remains a promising strategy for addressing and potentially overcoming MDR. In a se...

Circumvention of multi-drug resistance of cancer cells by Chinese herbal medicines

Multi-drug resistance (MDR) of cancer cells severely limits therapeutic outcomes. A proposed mechanism for MDR involves the efflux of anti-cancer drugs from cancer cells, primarily mediated by ATP-binding cassette (ABC) membrane transporters including P-glycoprotein. This article reviews the recent progress of using active ingredients, extracts and formulae from Chinese medicine (CM) in circumv...
